Target Information
Nitric oxide synthase (NOS) activity requires homodimerization as well as three cosubstrates (L-arginine, NADPH, and O₂) and five cofactors or prosthetic groups (FAD, FMN, calmodulin, tetrahydrobiopterin, and heme).
Three distinct NOS isoforms have been identified, encoded by separate genes:
- NOS1 (nNOS): Neuronal form, Ca²⁺/CaM-dependent
- NOS3 (eNOS): Endothelial form, Ca²⁺/CaM-dependent
These isoforms play key roles in nitric oxide signaling across various tissues.
