Product Specific Information

The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of the T and B lineage and activated mature T and B lymphocytes.
By itself, CD25 binds IL-2 with low affinity, but associates with CD122 and CD132 to form a high-affinity IL-2 receptor. Binding of IL-2 to both classes of IL-2 receptor is inhibited by PC61.5.
CD25 plays a role in lymphocyte differentiation and activation/proliferation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Mouse splenocytes, ≤ 0.25 µg/test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test.
Careful titration recommended for optimal performance.

Alexa Fluor™ 700 emits at 723 nm and is excited with a red laser (633 nm).
Recommended filters: 685 LP mirror and 710/20 filter.

Target Information

CD25 (IL2 receptor alpha chain / IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation.
The IL2 receptor complex (IL2RA, IL2RB, IL2RG) forms a high-affinity receptor; IL2RA homodimers form low-affinity receptors.
IL2 is produced mainly by mature T cells and regulates growth, differentiation, and cell death.
Mutations in IL2RA are associated with interleukin 2 receptor alpha deficiency and immune dysregulation.
