Product Specific Information

The YB5.B8 monoclonal antibody reacts with human CD117, also known as c-Kit, Steel factor receptor and stem cell factor receptor.
This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cell subsets and mast cells. Interaction of c-Kit and Steel factor promotes proliferation and differentiation of hematopoietic progenitor cells and mast cell differentiation. CD117 is also expressed by melanocytes and plays a role in their signaling and activation.

When staining hematopoietic stem cells (HSCs) from peripheral blood, clone 104D2 is recommended. See Product #17-1178.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on TF-1 human erythroleukemia cell line at ≤1 µg/test (100 µL final volume). Optimal performance requires empirical titration.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

KIT (c-KIT) is a proto-oncogene and a type 3 transmembrane receptor for mast cell growth factor (MGF, stem cell factor).
KIT regulates growth and activation of various hematopoietic and non-hematopoietic cells. Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism.
Activation by SCF leads to receptor dimerization and autophosphorylation of tyrosine residues, enabling downstream signaling via Ras/ERK, PI3K, PLC-γ, and JAK/STAT pathways.
c-Kit receptor activation is essential for hematopoiesis, stem cell maintenance, and gametogenesis.
