Product Specific Information

This MIH64 monoclonal antibody recognizes mouse VISTA (B7-H5, PD-H1).
It is validated for use in flow cytometry on both native and paraformaldehyde-fixed cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes (≤ 0.4 µg/test).
A test is defined as staining a 100 µL cell sample (10^5–10^8 cells/test).
Antibody titration is recommended for optimal performance.

Compatibility Notes:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer:
Updated formulation for optimal NovaFluor staining.
Add 5 µL per sample to antibody cocktails before combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10^3–10^8 cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.
Excitation: 552 nm; Emission: 612 nm; Laser: 561 nm (Yellow).

Note:
NovaFluor Yellow 610 may non-specifically stain B cells in SJL mice.
No non-specific staining observed in BALB/c or C57BL/6 mice.

VISTA (V-domain Ig containing suppressor of T-cell activation)
Also known as B7-H5 or PD-1H, encoded by the VSIR gene.

• Expressed mainly on hematopoietic cells (myeloid > T cells).
• Absent on B cells.
• Suppresses CD4/CD8 T-cell proliferation and cytokine production.
• Potential target for cancer immunotherapy.
• Known ligands: CD28H, VSIG-3, VSIG-8.
