Product Specific Information

The 9F10 monoclonal antibody reacts with human CD49d, the 150 kDa integrin alpha4 subunit. CD49d forms a complex with integrin beta1 (CD29), known as VLA-4, which serves as a receptor for fibronectin and VCAM-1 (CD106). This complex is expressed by thymocytes, peripheral lymphocytes, monocytes, and eosinophils. CD49d also associates with integrin beta7 and binds to MadCAM-1.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells at 5 µL (0.125 µg) per test.
A test is defined as the antibody amount required to stain a cell sample in a final volume of 100 µL (typically 10⁵–10⁸ cells/test).

Target Information

Integrin alpha 4 (CD49d, ITGA4) is part of the integrin alpha chain family. Integrins are heterodimeric membrane proteins composed of alpha and beta chains. The alpha4 chain associates with either beta1 or beta7. Ligand-binding sites for both integrin α4β1 and α4β7 are located on the alpha4 chain, binding to MadCAM, VCAM, and fibronectin. MadCAM is the principal ligand for α4β7. Notably, HIV-1 envelope can mimic MadCAM by binding and signaling through integrin α4β7, a gut mucosal homing receptor for peripheral T cells.
