Target Information

Ephrin A5 is a cell surface GPI-bound ligand for Eph receptors, a family of receptor tyrosine kinases crucial for migration, repulsion, and adhesion during neuronal, vascular, and epithelial development. It binds promiscuously to Eph receptors on adjacent cells, leading to contact-dependent bidirectional signaling.
Forward signaling occurs downstream of the receptor, while reverse signaling occurs downstream of the ephrin ligand. Ephrin A5 induces compartmentalized signaling within caveolae-like membrane microdomains, requiring Fyn tyrosine kinase activity.
It activates EPHA3 to regulate cell-cell adhesion and cytoskeletal organization, and with EPHA2 may regulate lens fiber cell shape and transparency. Ephrin A5 also mediates axon fasciculation and communication between pancreatic islet cells, influencing glucose-stimulated insulin secretion. It is a cognate/functional ligand for EPHA7, involved in brain development and modulation of cell-cell adhesion and repulsion.
