Target Information
Programmed death-ligand 2 (PD-L2), also known as B7-DC, belongs to the B7 ligand family within the immunoglobulin superfamily and acts as a ligand for programmed cell death protein 1 (PD-1), along with PD-L1. PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Its expression is upregulated by Th2 cytokine IL-4 and to a lesser extent by Th1 cytokines TNF-alpha and IFN-gamma.
PD-L2 exhibits 2–6 times higher affinity to PD-1 compared to PD-L1 and functions as an inhibitory immune checkpoint molecule, regulating T cell activation, proliferation, and cytokine secretion. Interestingly, PD-L2 can also stimulate T cell proliferation and cytokine production independently of PD-1, suggesting additional receptor interactions.
Recent studies indicate PD-L2 binds to repulsive guidance molecule b (RGMb), which influences respiratory tolerance development in the lung through BMP and/or neogenin signaling pathways. The human PD-L2 monomer consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
