Target Information

Nectin-3 is an 83 kDa type I transmembrane glycoprotein, originally identified as a poliovirus receptor-related protein (PRR). It belongs to the immunoglobulin superfamily and functions as a calcium-independent cell–cell adhesion molecule. The nectin family (nectin-1 to -4) mediates homophilic and heterophilic interactions to form various cell junctions, including cadherin-based adherens junctions in epithelial cells, synaptic junctions in neurons, and Sertoli cell–spermatid junctions in the testis.

Nectin-2 and Nectin-3 are widely expressed, while Nectin-1 is primarily found in the brain. Nectin-3 has three isoforms (α, β, γ), with Nectin-3α being the largest. It is predominantly expressed in testis and placenta, as well as in many cell lines. Nectin-3 interacts with afadin, an actin-binding protein, at cadherin-based junctions, and its ectodomain binds strongly to CD155 (Poliovirus Receptor). Experimental antibody injection into seminiferous tubules can disrupt actin filaments in Sertoli cells, leading to spermatid exfoliation.
