Product Specific Information

The A2MR-a2 monoclonal antibody reacts with human CD91 (also known as LRP-1 or alpha-2 macroglobulin receptor).
CD91 is a 600 kDa type I transmembrane protein composed of alpha and beta chains. The alpha chain binds diverse ligands, while the beta chain contains tyrosine phosphorylation motifs that interact with adaptor and signaling proteins. CD91 mediates ligand endocytosis and lysosomal trafficking.

This receptor is expressed by monocytes, dendritic cells, hepatocytes, fibroblasts, adipocytes, neurons, and syncytiotrophoblasts. It plays roles in antigen cross-presentation (via MHC class I), cell migration, and activation of signaling cascades.

• Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells
  • Recommended use: 5 µL (0.06 µg) per test in 100 µL final volume
  • Cell number: 10⁵–10⁸ cells/test (empirically determined)

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm).
Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
Ensure instrument compatibility for detection.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)
  Clone-specific performance after fixation should be verified empirically.

Filtration: 0.2 µm post-manufacturing filtered

Target Information

The encoded protein is an endocytic receptor involved in intracellular signaling, lipid homeostasis, and clearance of apoptotic cells.
It is essential for A2M-mediated clearance of secreted amyloid precursor protein and beta-amyloid, the major component of amyloid plaques in Alzheimer’s disease. Expression decreases with age and is reduced in Alzheimer’s patient brain tissue.
