Product Specific Information

Description:
The M1/69 monoclonal antibody reacts with the mouse CD24 molecule (Heat Stable Antigen, HSA). This 35–50 kDa GPI-anchored membrane protein is expressed on erythrocytes, thymocytes, peripheral lymphocytes, and myeloid cells. CD24 expression varies across lineages and is used to distinguish B lymphopoiesis stages in mouse bone marrow. CD24 also binds P-selectin (CD62P).

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes. Recommended ≤0.5 µg/test in 100 µL final volume (10⁵–10⁸ cells/test). Optimize titration for best performance.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by a violet laser (405 nm) and emits at 645 nm. Recommended filter: 660/20 bandpass. Ensure instrument compatibility.

Usage Recommendations:

• When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.
• Protect from light due to photo-induced oxidation sensitivity.
• For fixation, use IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333). Samples can be stored up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at various stages of B-cell development—from pro-B cells in bone marrow to mature surface Ig-positive B cells. Expression is low or absent in plasma cells. CD24 is also found in B-lineage leukemias and lymphomas, and may regulate B-cell proliferation, maturation, and autoimmunity control.
