Product Specific Information

Description:
The eBio64DEC17 antibody reacts with human IL-17A and is a neutralizing antibody. IL-17A is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in autoimmune and inflammatory diseases such as rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.

Human IL-17A exists as a glycosylated 20–30 kDa homodimer and induces secretion of IL-6, IL-8, G-CSF, and PGE2 in fibroblasts and epithelial cells. It also enhances ICAM-1 expression and supports hematopoietic progenitor proliferation.

Applications Reported:
For use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by intracellular staining of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended use: 5 µL (1.0 µg) per test in 100 µL final volume.

Fluorochrome Information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm and is optimized for cytometers with a 355 nm UV laser. Protect from light to prevent photo-induced oxidation.

Fixation Compatibility:
Compatible with short-term methanol-based fixation; avoid storage in methanol-containing buffers for over 1 hour. Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75).

Target Information

Interleukin-17A (IL-17A, CTLA-8) is a 32 kDa disulfide-linked homodimer cytokine produced by CD4+ T cells. It regulates inflammatory responses, enhances ICAM-1 expression, and promotes secretion of IL-6, IL-8, G-CSF, and PGE2. IL-17A signals through IL-17R/CDw217 and activates NF-κB and MAPK pathways, contributing to immune regulation and inflammation.
