Protein kinases are enzymes that transfer a phosphate group from ATP to an acceptor amino acid in substrate proteins. This mechanism mediates most signal transduction in eukaryotic cells, regulating metabolism, transcription, cell cycle, cytoskeletal rearrangement, movement, apoptosis, and differentiation.
The protein kinase family includes over 500 gene products, classified into 8 major groups based on catalytic domain sequence comparison. The STE group (homologs of yeast Sterile 7, 11, 20 kinases) consists of 50 kinases related to MAPK cascade families (Ste7/MAP2K, Ste11/MAP3K, Ste20/MAP4K). MAP kinase cascades, including MAPK and upstream kinases (MAPKKs), are well characterized in the yeast pheromone response pathway.
