Target Information
NUAK1 (ARK5) is a serine/threonine kinase involved in multiple cellular processes. Overexpression of this kinase is associated with tumor invasion and metastasis, particularly in colorectal cancer. It regulates cellular senescence and ploidy by phosphorylating LATS1 at Ser-464, controlling its stability. ARK5 also influences cell adhesion through regulation of the myosin protein phosphatase 1 (PP1) complex by phosphorylating PPP1R12A, which interacts with 14-3-3 proteins to modulate myosin MLC2 dephosphorylation.
Additionally, ARK5 may participate in DNA damage response by phosphorylating p53/TP53 at Ser-15 and Ser-392 and contributing to transcriptional activation of CDKN1A/WAF1. It may promote tumor invasion and metastasis under AKT1 regulation and suppress Fas-induced apoptosis by phosphorylating CASP6. ARK5 also plays a role in UV-induced DNA damage response by phosphorylating CDKN1A in association with STK11, facilitating its degradation for efficient DNA repair.
