Target Information
Mitochondrial transcription factor A (TFAM) is a nuclear-encoded protein that is imported into mitochondria, where it plays a key role in maintaining and expressing mitochondrial DNA (mtDNA). TFAM regulates mtDNA replication and transcription, ensuring proper mitochondrial function. It contains an LC3 interacting region (LIR) motif that allows binding to autophagy proteins such as LC3, facilitating the removal of leaked mtDNA and limiting inflammation. TFAM levels differ across tissues and influence oxidative phosphorylation (OXPHOS) capacity. Disruptions in TFAM function are linked to mitochondrial dysfunction and diseases associated with mtDNA stress.
