Target Information
HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under reduced oxygen tension. HIF-1 is a heterodimer composed of alpha and beta subunits, both belonging to the PAS family of transcription factors. It regulates the adaptive response to hypoxia by activating transcription of genes such as erythropoietin, glucose transporters, glycolytic enzymes, and VEGF.
HIF1-alpha influences hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. It interacts with p53 under hypoxic conditions to regulate its own expression via MDM2-mediated ubiquitination. Additionally, HIF1-alpha induction of NOX4 helps maintain ROS levels and supports hypoxia-induced proliferation.
In humans, HIF1A is located on chromosome 14q and its C-terminal region binds to p300, forming complexes that induce hypoxia-responsive genes. Hypoxia plays a key role in diseases such as ischemia, cancer, pulmonary hypertension, congenital heart disease, and COPD.
