Target Information

L-glutamate is the major excitatory neurotransmitter in the central nervous system, activating both ionotropic and metabotropic glutamate receptors. Glutamatergic neurotransmission is essential for normal brain function and is implicated in various neuropathologic conditions.

Metabotropic glutamate receptors (mGluRs) are G protein-coupled receptors divided into three groups based on sequence homology, signaling mechanisms, and pharmacologic properties:

• Group I: GRM1, GRM5 – activate phospholipase C
• Group II: GRM2, GRM3
• Group III: GRM4, GRM6, GRM7, GRM8

Groups II and III are linked to inhibition of the cyclic AMP cascade, differing in agonist selectivity. Multiple transcript variants encoding different isoforms have been identified for this gene.
