Target Information
TRAFs are signal transducers for members of the TNFR superfamily, critically involved in activation of MAP3K family members in NF-κB and JNK signaling and apoptosis.
TRAF7, a novel 670 amino acid protein, is the human homolog of mouse Traf7 and consists of:
- N-terminal RING finger domain
- Central TRAF domain
- Coiled-coil motif
- Seven WD40 repeats at the C-terminus
It has E3-ubiquitin ligase activity for self-ubiquitination and specifically interacts with MEKK3 to mediate MEKK3-induced AP1 and CHOP activation, inducing apoptosis through Caspase-independent pathways.
TRAF7 is multifunctional, involved in sumoylation and negative regulation of c-Myb, thereby modulating proliferation and differentiation of hematopoietic cells.
