Target Information
Ago 단백질은 체세포에 널리 발현되며, miRNA와 결합하여 RNA 침묵 경로에서 핵심 역할을 합니다. 그중 Ago2 단백질만이 endonucleolytic 또는 “Slicer” 활성을 가지며, miRNA와 완전 상보적인 mRNA를 절단할 수 있습니다. 부분 상보적인 경우 절단 대신 번역 억제를 유도합니다. Ago2는 RNA 간섭, 번역 억제, 이질염색질화 등 다양한 small RNA 유도 유전자 침묵 과정에서 중요한 역할을 하며, RISC 복합체의 핵심 요소로서 siRNA 또는 miRNA에 의해 유도된 표적 mRNA의 분해를 촉진합니다. 또한 Dicer 비의존적 RNA 절단 및 miRNA 성숙 조절에도 관여합니다. Ago2의 과발현은 암세포 성장 및 생존율과 연관되어 있으며, 마우스 모델에서 Ago2 유전자의 결손은 배아 발달 및 B-림프구, 적혈구 발달의 핵심 조절자로서 필수적임이 밝혀졌습니다.
