ETS1 is a 441 amino acid DNA-binding protein belonging to the ETS family. It contains an N-terminal pointed domain, a central transactivation domain, and two auto-inhibitory domains flanking the conserved C-terminal ETS domain. ETS1 interacts with transcription factors such as PEBP2alphaA and p300/CBP, mediating transactivation of genes including Osteopontin, Presenilin-1, Stromelysin, and Collagenase. Activated by Ras/MAPK signaling, ETS1 expression increases in resting T-cells and is regulated by factors such as AP-1, Sp-1, c-myb, and MafB. It is also activated by Sp100 in nuclear bodies and interacts with EAP1/Daxx, STAT6, and GATA3, influencing gene expression including MMP1, BCL2, Socs-1, and IL-5.
