Target Information
SMCR8 is a component of the C9orf72–SMCR8 complex, which exhibits guanine nucleotide exchange factor (GEF) activity and regulates autophagy. Within the complex, C9orf72 and SMCR8 likely serve as catalytic subunits that facilitate the exchange of GDP to GTP, activating RAB8A and RAB39B to promote autophagosome maturation.
Additionally, the complex negatively regulates autophagy initiation by interacting with the ATG1/ULK1 kinase complex and inhibiting its kinase activity.
SMCR8 also modulates mTORC1 signaling by promoting phosphorylation of mTORC1 substrates. Beyond its cytoplasmic role, SMCR8 localizes to the nucleus, associates with chromatin, and downregulates expression of ULK1 and WIPI2 genes.
