Target Information
The 26S proteasome is a multicatalytic proteinase complex composed of two major complexes: a 20S core and a 19S regulator.
- The 20S core consists of four rings of 28 non-identical subunits (two alpha and two beta rings).
- The 19S regulator includes a base (with ATPase and non-ATPase subunits) and a lid (up to 10 non-ATPase subunits).
Proteasomes are abundant in eukaryotic cells and degrade peptides in an ATP/ubiquitin-dependent, non-lysosomal pathway. The immunoproteasome variant processes class I MHC peptides.
The PSMD2 gene encodes a non-ATPase subunit of the 19S regulator lid, potentially involved in TNF signaling via interaction with the TNF type 1 receptor. A pseudogene exists on chromosome 1.
