Target Information
The PKC family of serine/threonine kinases, including PRKCI (PKC iota), is activated intracellularly by signal transduction pathways. In humans, at least 12 PKC isoforms have been identified, differing in structure, localization, and substrate specificity. Conventional PKC members (alpha, beta I, beta II, gamma) are Ca²⁺- and phospholipid-dependent, while PRKCI is calcium-independent and phospholipid-dependent. It is not activated by phorbol esters or diacylglycerol.
PRKCI interacts with the small GTPase RAB2 at vesicle tubular clusters (VTCs), phosphorylating GAPDH and influencing microtubule dynamics in the early secretory pathway. It plays a critical role in BCL-ABL-mediated resistance to apoptosis, protecting leukemia cells against drug-induced apoptosis. A single exon pseudogene is mapped on chromosome X.
