Background

  • Domain: The LXXLL motif is a coregulator signature essential for transcriptional corepression.
  • Function: Acts as an E3-type SUMO ligase, stabilizing interactions between UBE2I and substrates, and serves as a SUMO-tethering factor. Plays a key role in transcriptional coregulation in pathways such as STAT, p53, Wnt, and steroid hormone signaling. Promotes PARK7 sumoylation.
  • Pathway: Protein modification; protein sumoylation.
  • PTM: Sumoylated at Lys-35, required for TCF4 sumoylation and transcriptional activation. Represses LEF1 transcriptional activity. SUMO1 is the preferred conjugate.
  • Similarity: Belongs to the PIAS family; contains one SAP domain and one SP-RING-type zinc finger.
  • Subunit: Interacts with AR, GATA2, LEF1, TP53, and STAT1 (IFNG-induced). Binds AT-rich DNA sequences (MARs/SARs). Interacts with TICAM1 and KLF8, leading to SUMO ligation and repression of KLF8 transcriptional activity and cell cycle progression.
  • Tissue Specificity: Highly expressed in testis; lower levels in spleen, prostate, ovary, colon, and peripheral blood leukocytes.
