Background
Activating transcription factor 1 (ATF1) is a member of the ATF subfamily and the bZIP (basic-region leucine zipper) family. It regulates the expression of downstream target genes involved in growth, survival, and other cellular activities. The protein is phosphorylated at serine 63 by several kinases, including cAMP-dependent protein kinase A, calmodulin-dependent kinase I/II, mitogen- and stress-activated protein kinase, and cyclin-dependent kinase 3 (cdk-3). Phosphorylation enhances its transactivation and transcriptional activities, promoting cell transformation. Chromosomal translocations involving ATF1 and FUS (chromosome 16) or EWSR1 (chromosome 22) generate chimeric proteins implicated in angiomatoid fibrous histiocytoma and clear cell sarcoma. A pseudogene also exists on another chromosome.
