Background
Class II major histocompatibility complex transactivator (CIITA) is a human protein containing an acidic transcriptional activation domain, four leucine-rich repeats (LRRs), and a GTP-binding domain. It functions as a positive regulator of MHC class II gene transcription and is known as the "master control factor" for their expression. CIITA binds GTP to facilitate nuclear transport but does not directly bind DNA. Instead, it exhibits intrinsic acetyltransferase (AT) activity, acting in a coactivator-like manner. Mutations in this gene are associated with bare lymphocyte syndrome type II (hereditary MHC class II deficiency), and may contribute to susceptibility to rheumatoid arthritis, multiple sclerosis, and myocarditis.
