Target Information
HMGB1 (High-mobility group box-1) 단백질은 원래 비히스톤 DNA 결합 핵 단백질로 알려졌습니다. 최근 연구에 따르면 손상되거나 괴사된 세포에서 HMGB1이 세포외로 방출되어 염증성 사이토카인으로 작용합니다.
마우스 HMGB1은 215 아미노산으로 구성된 단일 사슬 폴리펩타이드이며, 두 개의 양전하 DNA 결합 도메인(HMG box A: aa 9–79, box B: aa 89–162)과 음전하의 C-말단 산성 꼬리 영역(30 aa)을 포함합니다.
Residues 28–44 및 180–185는 핵 내 이동 신호(NLS)를 포함하며, B box는 사이토카인 활성을, A box는 전사인자의 helix-loop-helix 도메인과 연관되어 있습니다.
HMGB1은 염증 매개체로서 단핵구 이동과 사이토카인 분비를 촉진하고, T세포-수지상세포 상호작용을 매개합니다. RAGE 및 TLR2, TLR4 수용체를 통해 세포 신호를 전달하며, 대부분의 세포핵과 세포질에 존재하는 고도로 보존된 단백질입니다.
또한 HMGB1은 무균 염증의 초기 유발자로서, 외상, 패혈증, 허혈-재관류 손상 등 다양한 병리적 상황에서 발현이 증가하는 것으로 보고되었습니다.
