Product Specific Information

This F38-2E2 monoclonal antibody reacts with human CD366 (TIM3, HAVCR2), a cell surface receptor expressed on activated CD4⁺ T cell subsets (Th1, Th17, Treg), CD8⁺ T cells, monocytes, dendritic cells, and mast cells.
CD366 exists in membrane-bound and soluble forms due to alternative splicing, with Galectin-9 as its ligand. It negatively regulates CD4⁺ T cells and influences Th1/Th17 cytokine secretion. TIM3 is implicated in immune tolerance, autoimmune diseases (e.g., multiple sclerosis), and HIV infection.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry of stimulated human peripheral blood cells. Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Alexa Fluor 700 emits at 723 nm and is excited by the red laser (633–647 nm). Instruments typically require a 685 LP mirror and 710/20 band pass filter.

Target Information

TIM3 (HAVCR2) is a 281-amino-acid, type I transmembrane glycoprotein expressed on terminally differentiated CD4⁺ Th1 and CD8⁺ Tc1 cells. It includes an IgV-like domain, a mucin-like extracellular domain, and a conserved cytoplasmic tyrosine phosphorylation motif.
TIM3 is involved in macrophage activation, autoimmune disease induction, and immune tolerance regulation. It down-regulates Th1-mediated immune responses and has been linked to experimental autoimmune encephalomyelitis (EAE) and immune pathogenesis.
