Identify and characterize palmitoylated proteins with Click-iT® Palmitic Acid, Azide using powerful click chemistry — a simple and robust two-step labeling and detection technique.

1. Labeling Step: The azide-containing biomolecule is fed to cells or animals and incorporated into proteins.
   • The small azide tag allows normal enzymatic incorporation, unlike bulky biotin or fluorescent dyes.
2. Detection Step: Detection utilizes chemoselective ligation (“click” reaction) between an azide and an alkyne.
   • Modified proteins are detected with alkyne-containing dyes or haptens using either the Click-iT® Cell Reaction Buffer Kit or the Click-iT® Protein Buffer Kit.

Application

• Cell-based analysis: Fluorescence microscopy, flow cytometry, or high-content imaging (HCS) with other biomarkers for comprehensive results.
• Protein-based detection: Achieve femtomole-level sensitivity in 1-D gels and western blots, or perform LC-MS/MS and MALDI-MS analysis.
