Target Information
Janus kinase 3 (JAK3) phosphorylates members of the STAT (signal transduction and activators of transcription) family. JAK3 is predominantly expressed in immune cells and transduces signals in response to activation of interleukin receptors.
Upon stimulation by cytokines or related factors (e.g., interferon, interleukins), JAKs are activated via phosphorylation at two adjacent tyrosine residues. This activation leads to phosphorylation of STAT proteins, which then dimerize and translocate to the nucleus to regulate transcription of various genes, including interferon-stimulated genes.
JAK2 is essential for IFN gamma-receptor complex initiation, while JAK1 functions as an amplifier. Some studies suggest that Tyk2 and JAK3 may substitute for JAK2 if properly positioned within the receptor complex.
