Tumor necrosis factor (TNF) receptor-associated factors (TRAFs) act as adaptor proteins linking the TNF receptor superfamily to signaling pathways, playing key roles in regulating cell death and stress responses.
TRAF proteins share a homology region that facilitates binding to cell receptors and other TRAF proteins, activating various signaling cascades.
TRAF2 and TRAF3 directly bind to CD40, an NF receptor superfamily member, and are essential for NF-kappa-B activation in mouse B lymphocytes.
TRAF2, together with TRAF6, is required for CD40 signaling in non-hematopoietic cells and interacts with lymphotoxin-beta receptor (LTbetaR) along with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
