Target Information
Granzyme B is a member of the granzyme serine protease family, found in the granules of cytotoxic T cells and NK cells. It has been described as CGL1 (cathepsin G-like-1) and CTLA-1 (cytotoxic T lymphocyte-associated serine esterase 1). Granzyme B plays a key role in apoptosis induction via cytotoxic T cell interaction, involving the mannose 6-phosphate receptor as a death receptor. It cleaves caspase-3, initiating the caspase cascade leading to DNA fragmentation and apoptosis, and can also act through mitochondrial pathways by cleaving Bid protein. Granzymes are neutral serine proteases stored in specialized lytic granules of CTLs and NK cells. Several granzymes (A–G) have been cloned from mouse CTLs and NK cells, while fewer have been identified in humans.
