Target Information

The tachykinins are a family of small peptides that include the neurotransmitters substance P, neurokinin A, and neurokinin B, acting on three related but distinct seven transmembrane G-protein-coupled receptors. The NK3 receptor (NK3R) shows the greatest affinity for neurokinin B and is highly expressed in the supraoptic and paraventricular nuclei. Upon ligand binding, NK3R activates a phosphatidylinositol-calcium second messenger system, likely leading to the release of vasopressin and oxytocin into circulation. NK3R may be involved in learning and memory, as mice lacking this gene display cognitive deficits. Although NK3R has been suggested to regulate vagal afferent relay neurons, activation in airway nerves is more likely mediated by substance P or neurokinin A, since these nerves do not express neurokinin B.
