Target Information

TAK1 (also known as MAP3K7) is a serine/threonine kinase with multiple signaling functions. Initially identified as a TGF-β–activated kinase, TAK1 interacts with TGF-β receptor and TRAF6 to modulate activation of JNK and p38. It also participates in Wnt, BMP, activin, and other signaling pathways responding to various stimuli such as thyroid hormone and osmotic stress. TAK1 regulates wound healing via activation of von Hippel-Lindau tumor suppressor expression and represses PDGF-B and integrins. Additionally, TAK1 is essential for IKK activation in pathways including IL-1, IL-6, IL-18, TNF, CD40, TLR, and RIG-I, involving TAB2/3 and ubiquitination. TAK1 also represses human telomerase reverse transcriptase, suggesting a role in cell lifespan regulation.
