Target Information

PYCARD functions as a key mediator in apoptosis and inflammation. It promotes caspase-mediated apoptosis involving caspase-8 and caspase-9, and is involved in activation of the mitochondrial apoptotic pathway. PYCARD mediates macrophage pyroptosis, forming the ASC pyroptosome upon potassium depletion and activating caspase-1. It acts as an adapter in inflammasome assembly, regulating processing and secretion of proinflammatory cytokines through pyrin and CARD domains. It is required for recruitment of caspase-1 to inflammasomes containing NLRP2, NLRP3, AIM2, and IFI16, and facilitates processing of procaspase-1 in NLRP1 and NLRC4 inflammasomes. Additionally, PYCARD participates in NOD2-related inflammasome activation and may regulate NF-κB signaling. Isoforms 2 and 3 have distinct regulatory roles in inflammasome function. The protein also contributes to adaptive immune responses, dendritic cell maturation, and cytokine transcriptional activation.
