Proteolytic degradation is essential for maintaining appropriate levels of short-lived and regulatory proteins involved in metabolism, stress response, antigen presentation, receptor modulation, cell cycle regulation, transcription, and signaling.
The ubiquitin-proteasome pathway deconstructs most cytosolic and nuclear proteins in eukaryotic cells, while other proteins are degraded via the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum.
The 26S proteasome is an ATP-dependent, multisubunit (~31) complex with an approximate molecular weight of 2.5 MDa. It consists of a 20S proteolytic core complex capped by 19S regulatory subunits that recognize ubiquitinated proteins and facilitate unfolding and translocation into the 20S core.
The PA28/11S REG Activator complex (α, β, γ subunits) enhances 20S core activity.
Defects in the ubiquitin-proteasome pathway are linked to several genetic diseases, including cystic fibrosis (CFTR), Angelman’s syndrome (E6-AP), and Liddle syndrome (ENaC).
