Target Information

Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor involved in the detection of viral double-stranded RNA (dsRNA). Together with MDA5, RIG-I activates the innate immune response by recognizing viral RNA.
RIG-I is activated by short dsRNAs lacking a 5'-triphosphate end, whereas MDA5 responds to long dsRNAs. Upon activation, both signal through IPS-1 to activate NF-κB and IRF-3, leading to apoptosis, cytokine signaling, and inflammation.
RIG-I is essential for antiviral signaling against influenza A/B, RSV, paramyxoviruses, Japanese encephalitis virus, and West Nile virus.
MicroRNA-146a negatively regulates RIG-I-dependent antiviral responses by targeting TRAF6, IRAK1, and IRAK2.
Recent evidence also suggests RIG-I detects cytosolic DNA through RNA polymerase III activity.
