Target Information

G protein-coupled receptor GPR49 (LGR5) is expressed in brain, skeletal muscle, placenta, and spinal cord. ESTs have been isolated from normal human brain (amygdala), embryo, and placenta libraries, as well as from human germ cell, uterus, and brain libraries.

G-protein Coupled Receptors (GPCRs) represent one of the largest families of signaling molecules, with over a thousand predicted members. They share a structural motif of seven membrane-spanning helices and exist in both active and inactive forms. Ligand binding induces conformational changes that activate G-proteins and downstream signaling pathways.

Regulatory mechanisms that “turn off” GPCR signaling are crucial for cellular physiology and are key targets in pharmacological research. Notably, GPCR-targeted drugs such as Zyprexa (Eli Lilly) and Claritin (Schering-Plough) have achieved significant success in mental health and allergy treatment markets.
