Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It acts as a receptor for the hemoglobin-haptoglobin complex and helps protect tissues from oxidative damage caused by free hemoglobin. CD163 is involved in iron recycling through endocytosis of hemoglobin/haptoglobin complexes and subsequent heme breakdown.

CD163 is expressed on human monocytes and macrophages, particularly in the late phase of inflammation. It is found on all circulating monocytes and most tissue macrophages, except those in mantle zones, germinal centers, interdigitating reticulum cells, and Langerhans cells. Expression is high on CD14⁺ and CD16⁺ monocytes and is upregulated by IL-10, IL-6, and dexamethasone. LPS and PMA stimulate shedding of CD163 from the cell surface into plasma or supernatant.

CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for multimeric haptoglobin (HP1F phenotype) than for dimeric haptoglobin (HP1S phenotype). It also triggers intracellular signaling cascades involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL6 and CSF1.
