Target Information
Specialized cells at the midline, which separates the left and right halves of the central nervous system, play key roles in directing growth cone behavior. In vertebrates and invertebrates, midline cells produce guidance cues such as nectins and slit, acting as attractants and repellents. Robo proteins serve as axon guidance receptors and belong to a conserved subfamily of Ig superfamily proteins. Robo1 acts as a receptor for the repellent Slit and functions cell-autonomously. Non-crossing axons express high levels of Robo, while crossing axons modulate expression before and after midline crossing. Robo1 and Robo2 are human homologs of the Drosophila Roundabout protein; Robo1 is also homologous to the C. elegans sax3 gene, and Robo2 to the zebrafish astray gene.
