Product Specific Information
The monoclonal antibodies AE1 and AE3 recognize many members of the acidic and basic cytokeratin family. Cytokeratins are intermediate filament proteins forming part of the cytoskeleton. They are expressed in epithelial cells and are developmentally regulated. Many tumors also express these proteins, and their detection aids in identifying the origin of neoplasms.
- AE3 antibody recognizes cytokeratin proteins of 65–67, 64, 59, 58, 56, 54, and 52 kDa (CK1–8).
- AE1 antibody recognizes cytokeratin proteins of 56.5, 54, 50, 48, and 40 kDa (CK10, 14, 15, 16, 19).
- Applicable to tissue samples from human, mouse, rat, primates, dog, cat, rabbit, and chicken.
Applications Reported:
Useful for immunohistochemical staining, immunocytochemistry, and staining of frozen or formalin-fixed paraffin-embedded tissue sections.
Applications Tested:
Validated for IHC of formalin-fixed paraffin-embedded tissue using high pH antigen retrieval. Recommended dilution: 2 µg/mL (optimize per assay).
Usage Note:
Centrifuge conjugate solution briefly before use and apply only the supernatant to reduce nonspecific background staining.
Target Information
Cytokeratin pan belongs to a subfamily of intermediate filament proteins with diverse biochemical properties, represented in epithelial tissues by at least 20 polypeptides (40–68 kDa, pI 4.9–7.8). Expression patterns vary by epithelial type and differentiation stage, serving as valuable markers in cancer research, tumor diagnosis, and cytopathology. Cytokeratin profiles assist in distinguishing epithelial malignancies and are useful in flow cytometric assays.
