Target Information
Transglutaminase (TGase)는 단백질 간 e-(γ-glutamyl) lysine isopeptide 결합을 형성하여 교차결합을 촉매하며, 활성에는 Ca²⁺ 결합이 필요합니다. 포유류에서는 8가지의 TGase 동종효소가 확인되었습니다. Tissue transglutaminase (tTGase, TGase 2)는 N-말단 β-sandwich, 촉매 코어, 두 개의 C-말단 β-barrel 도메인을 포함한 네 개의 도메인을 가지고 있습니다. tTGase는 세포사멸, 증식, 분화 및 수용체 매개 내포작용에 관여할 수 있습니다. 알츠하이머병 뇌에서는 Ab 펩타이드, β-amyloid 전구체 단백질, tau 단백질 등의 중합으로 인한 신경섬유 엉킴(neurofibrillary tangles) 형성과 관련된 tTGase 활성 증가가 관찰됩니다.
