Target Information
Calcineurin is a Ser/Thr, calcium and calmodulin-dependent protein phosphatase essential for T cell activation. It consists of two subunits:
- Calcineurin A (CnA): 60 kDa catalytic subunit
- Calcineurin B (CnB): 19 kDa regulatory subunit containing four EF-hand calcium-binding sites
Calcineurin dephosphorylates the nuclear factor of activated T-cells (NF-AT), enabling its activation and nuclear translocation. Immunosuppressive drugs such as cyclosporin A and FK506 inhibit this process by forming complexes that block substrate access to the catalytic domain of CnA without affecting phosphatase activity.
