Target Information

PKN2 (PRK2), a close relative of the PKCs, is a regulator of mitosis and cytokinesis exit. It plays a key role in:

• Cell cycle progression and actin cytoskeleton assembly
• Cell migration, adhesion, and tumor cell invasion
• Transcription activation signaling processes

It phosphorylates CTTN in hyaluronan-induced astrocytes, reducing its ability to associate with filamentous actin.
PKN2 also phosphorylates HDAC5, impairing its nuclear import, and acts as a direct RhoA target regulating junction maturation in epithelial cells.
It is essential for G2/M phase progression and abscission during cytokinesis (ECT2-dependent).
Additionally, PKN2 stimulates FYN kinase activity required for keratinocyte differentiation and regulates epithelial bladder cell migration and invasion.
It inhibits Akt pro-survival kinase activity and mediates Rho protein-induced transcriptional activation via the c-fos SRF pathway.
PKN2 phosphorylates HCV NS5B, stimulating HCV RNA replication.
