- Positive control: THP-1 cells treated with 80 nM TPA overnight, then 100 ng/mL LPS for 6 hours, and 300 ng/mL BFA for 3 hours (whole cell lysate).

Target Information
Interleukin-1 beta (IL-1 beta) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. IL-1 beta is synthesized in response to inflammatory stimuli as a 31 kDa inactive pro-form that accumulates in the cytosol. Cleavage of pro-IL-1 beta into the active 17 kDa protein requires activation of inflammasomes, multi-protein complexes that respond to pathogens and stress signals. Activated caspase-1 cleaves pro-IL-1 beta, enabling secretion via a non-classical pathway.
IL-1 beta signals through IL-1RI and IL-1RII receptors, shared with IL-1 alpha. Its activity is regulated by IL-1 Receptor Antagonist (IL-1RA), which blocks receptor binding. IL-1 beta plays a key role in innate immunity and inflammation, and elevated levels are linked to chronic inflammatory diseases. IL-1 beta neutralizing antibodies have potential therapeutic applications.
