Target Information
HIV-1 Tat (Trans-Activator of Transcription) consists of 86 to 101 amino acids depending on the subtype.
Tat is critical to HIV reproduction by compensating for a defect in its genome: the HIV RNA initially has a hairpin-structured portion which prevents full transcription.
A small number of RNA transcripts allow Tat protein production. Tat binds to and phosphorylates cellular factors, eliminating the hairpin RNA structure effect and enabling full transcription of HIV RNA.
This enhances transcription rate through a positive feedback cycle. The protein is released by infected cells and found in the blood of HIV-1 infected patients.
