Target Information
Apoptosis is an essential process for maintaining normal tissue homeostasis and function. One of the key regulatory pathways involves activation of the Fas receptor by its ligand, leading to formation of a Fas-associated death domain (FADD)-containing death-inducing signaling complex and activation of caspase-8. This enzyme subsequently activates downstream effector caspases such as caspase-3 and -7.
Overexpression of Toso (FAIM3), a novel regulator of Fas-induced apoptosis in lymphoid cells, has been shown to make Jurkat cells and transgenic mice resistant to Fas-induced apoptosis but not to TRAIL-induced apoptosis. Toso associates with FADD, suggesting it disrupts the formation of the death-inducing signaling complex.
