Target Information

The leptin (OB-R) receptor is a member of the cytokine receptor superfamily that plays an important role in mammalian body weight homeostasis and energy balance. Ligand-induced activation of OB-R by the adipose tissue-secreted hormone leptin triggers downstream signaling through the Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway. Leptin can homo-dimerize OB-R extracellular domains, initiating signaling via intracellular domain dimerization. Several isoforms of OB-R, differing in length and signaling capability, have been reported. RNA transcripts encoding the OB-R long form are found in the choroid plexus and hypothalamus, regions associated with satiety and energy expenditure control. Mutations in OB-R (db) or leptin (ob) genes in mice lead to early-onset obesity and related metabolic abnormalities such as hyperinsulinemia, insulin resistance, hyperglycemia, and increased adipose tissue.
