Product Specific Information

Description:
The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L, also known as LFA-1α. LFA-1 is a heterodimer composed of CD11a (αL) and CD18 (β2), functioning as an adhesion molecule critical for lymphocyte and granulocyte activity. Ligands include CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3). CD11a is expressed on all leukocytes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Validated for flow cytometry using mouse splenocytes. Recommended ≤ 0.4 µg/test (100 µL final volume). Cell density: 10⁵–10⁸ cells/test. Optimize titer for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample or per 100 µL of cell suspension.
  • Add buffer to antibody cocktails before mixing with cells.

NovaFluor conjugates use Phiton™ technology, employing nucleic acid dye structures engineered for optimal fluorescence with minimal spillover and spread.

Target Information

CD11a (LFA-1α, ITGAL):
CD11a pairs with CD18 to form LFA-1 (αLβ2 integrin), crucial for leukocyte adhesion via ICAM-1, -2, and -3. It mediates immune cell interactions, migration, and activation. Dysfunction is linked to disorders such as leukocyte adhesion deficiency and benign cephalic histiocytosis.
Therapeutic inhibition of CD11a (e.g., with Efalizumab) has clinical relevance in inflammatory disease modulation.
