Product Specific Information

The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L, also known as the lymphocyte function-associated antigen-1 (LFA-1) alpha chain.
LFA-1, formed by non-covalent association of CD11a with CD18 (integrin beta 2), serves as an important adhesion molecule involved in lymphocyte and granulocyte function.
CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3) are ligands for LFA-1. CD11a is expressed by all leukocytes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.4 µg/test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample before combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread.
Excitation: 509 nm; Emission: 665 nm; Laser: 488 nm (Blue).

Target Information

CD11a (LFA-1 alpha, ITGAL) with CD18 forms leukocyte function-associated antigen 1 (LFA-1), an αLβ2 integrin.
CD11a mediates leukocyte adhesion via ICAM-1, -2, -3 and participates in lymphocyte co-stimulatory signaling.
Expressed on all leukocytes, transitioning from low- to high-affinity conformation upon stimulation for tight ligand binding.
Involved in immune cell interaction, tissue-specific localization, differentiation, proliferation, and T-cell effector functions.
Therapeutic targeting of CD11a (e.g., Efalizumab) has applications in inflammatory disease treatment.
Two transcript variants encode different isoforms.
Associated diseases include benign cephalic histiocytosis and leukocyte adhesion deficiency.
