Target Information

The insulin-like growth factor-binding proteins (IGFBPs) are a family of homologous proteins that co-evolved with the IGFs. They act as shuttle molecules for soluble IGFs and regulate IGF signaling by influencing growth factor bioavailability and distribution. IGFBPs also exhibit IGF-independent biological activity.
Seven IGFBPs have been identified, each differing in tissue distribution, half-life, and modulation of IGF-receptor interactions. For example:

  • IGFBP1 is negatively regulated by insulin and highly expressed during fetal liver development.
  • IGFBP2 may function as a chaperone, expressed in fetal eye and brain.
  • IGFBP3 is the most abundant form, binding ~80% of serum IGFs.
  • IGFBP4 is released by dermal fibroblasts after incision injury.
  • IGFBP5 is secreted by myoblasts and may play a key role in muscle differentiation.
